When a big player wants to bet serious money....on a side bet, you could be looking at anything over 50dimes, the book might take the bet in pieces...adjusting the line/juice. It all depends on the game in question and the position of the book at the time of the bet.
As to the "millions" aspect....The largest bet that I have seen in the last year or so was $2million moneyline Colts vs Saints. That bet was taken at -200.

The line at the book in question (or in some cases, the company in question....see Sharpsticks posting), the line could change or, as I said in my post, it might change during the course of the bet...being broken down into multiple bets by the bettor that the book is accepting. Whether or not any other book in town would make a change based upon "anticipated" betting, would be up to the manager of that book.....
A bet at Caesars does not, necessarily, influence the line at The Mirage....the books are competing for business and might feel that their line will attract the money that it's looking for. A book may have too much money on one side and want to encourage money from the other side....
The Auburn situation today is a great example....read the article on the home page about how the game got pulled....guarantee that there was money all over town much earlier today that was "under the radar"....Books do not, neccessarily, care to share info....Mirage may have gotten hit late with one last bet; others may have already been hit.
