Damn, good question! There are so many funny ass comedians... Let's divide it between Old School and New School since comedy has changed so much between eras...
New School:
My personal favorite is Chris Rock...I could watch his comedy specials every day for a year and still laugh my ass off!
Honorable Mention goes to Dave Chappelle!!!
Old School:
This is tough...Even though I hate what he did with his career turning to Disney movies instead of hardcore comedy...Has to be Eddie Murphy...

the old Miller Lite commercials using retired sports stars were ALL very funny BUT the ones with Rodney were definately the best. Jay Leno uses his rapid fire style of delivery every night as part of his monolog and and it's usually the funnyist part.

But the criteria is SO varied-W.C. Fields ("I remember one time, we were stranded on a desert island. We lost our corkscrew, and had to survive on food and water for several days.....") was in vaudeville before he even made it to radio, and then movies (before tv, mind you) Milton Berle, Jack Benny, Sid Caesar.......same thing. Steve Allen, Johnny Carson, Jay Leno, Letterman......all RULED television. Does that make them the best? Being seen every night, would that make them the most popular?
Bob Newhart, Dick Van Dyke, Robin Williams, Ray Romano........all great standups that worked great on television
Then, we have the best comedians we've never heard of....
Couldn't. They worked "blue", which meant curse words were spoken, or word that were found "offensive"
